If you use a draft class, you won't play against them. It seems like draft classes don't even work in MP since I got the default 2k rookies. If you use a roster with the real rookies in there, you will play against the default 2k rookies during the Rookie Showcase, but it looks like that once they get drafted they're not on their teams, but the real rookies already on the roster are.
