I don't have any first hand experience running an NBA Live league but I would suggest that having a script to manage all your statistics is a must. Otherwise, you might want to have a few people working on the site to ensure that everything is kept up to date. You'll also need to work out what kind of league it will be (whether players will be competing online or whether they'll simply run teams while you simulate the results, etc) and devise some appropriate rules.
